Pharmacological Actions
Antimalarial Effect
It has a strong inhibitory effect on Plasmodium falciparum and other malaria parasites. It can effectively kill malaria parasites in the erythrocytic stage, thereby alleviating the symptoms of malaria and achieving the purpose of treating malaria.
Mechanism of Action
Similar to other artemisinin - based drugs, artesunate acts by generating free radicals after the cleavage of its peroxide bridge in the presence of iron ions in the malaria parasite. These free radicals alkylate and damage the proteins and nucleic acids of the malaria parasite, interfering with its normal metabolism and growth, and finally leading to the death of the parasite.
Clinical Applications
Treatment of Malaria
Artesunate is widely used in the treatment of various types of malaria, especially in the treatment of severe malaria. It can quickly reduce the parasite load in the blood, relieve the symptoms of malaria, and improve the prognosis of patients. In combination with other antimalarial drugs, it can achieve better curative effects and reduce the risk of drug resistance.
Prophylaxis of Malaria
In some malaria - endemic areas, artesunate can also be used for malaria prophylaxis, especially for people at high risk of malaria infection, such as travelers and field workers.
Dosage and Administration
The dosage of artesunate varies depending on the type and severity of malaria, as well as the age and body weight of the patient. For the treatment of uncomplicated malaria, the recommended oral dose is usually 4 - 6 mg/kg per day for 3 - 7 days. For severe malaria, the parenteral formulation (such as intravenous or intramuscular injection) is often used, and the initial dose is generally 2.4 mg/kg, followed by repeated doses according to the patient's condition.
